Using the standard curve method, the absolute quantity of RNA in test samples is measured. The standard curve graph on the left was created by performing 5-fold serial dilutions of cDNA generated from RNA extracted from a stock of Influenza A (IFV-A)/Puerto Rico/8/1934. The x-axis represents the log values of DNA starting dilution, while the y-axis indicates the measured quantification cycle (Cq) value. The graph on the right shows the amplification of cDNA made from RNA extracted from samples.
The amplification plot below was generated from Zika virus-containing samples. Samples were serially diluted 5-fold and gene amplification of the Zika envelop protein was observed. On the right, the melt curve plot indicates the melting temperature of a target. This specific melt curve shows high specificity to the Zika envelope protein gene, indicated by the peaks isolated to 78°C.
