Expression and purification of recombinant antibodies expressed in CHO cells

Cell media is collected, and the recombinant antibody, purified by protein A, is immobilized on a solid support. CHO cell media was loaded onto a protein A column, washed, and then eluted by low pH. Blue line is the A280. Antibody is eluted using a low pH buffer. The eluted antibody is immediately neutralized by high pH buffer. The antibody is then dialyzed into final storage buffer.

Analysis by SDS-PAGE and SEC-HPLC is performed on the purified antibody.

For SDS-PAGE, the purified antibody was electrophoresed on a reducing SDS-PAGE to separate heavy and light chains. MW=Protein Molecular Weight Markers. Lane 1 is Antibody at 1µg/lane. For SEC-HPLC, the purified antibody (50 µg) was loaded onto SEC-column. Elution profile shows 100% purity.
