This unique lethal model uses a non-mouse-adapted strain of DENV serotype 2 (D2Y98P), isolated from a DENV-infected patient in Singapore, which is highly infectious in AG129 mice.
The model provides consistent clinical manifestations of disease similar to those observed in human cases (Tan, et al., 2018). The DENV model has several advantages. Firstly, a low-challenge dose of the virus leads to transient asymptomatic infection followed by death a few days after viral clearance.
Secondly, it shows increased vascular permeability without morphological damage of the capillary endothelium, which is comparable to human disease. Finally, three vital pro-inflammatory cytokines implicated in Dengue Hemorrhagic Fever (DHF) are significantly elevated in D2Y98P-infected AG129 mice.
Description: Disease progression of dengue-2 D2Y98P and S221 strains in AG129 mice (n=15). DENV-2 D2Y98P was administered via the subcutaneousroute (SC), while S221 was inoculated
via the intravenous (IV) route).
Description:Serum collected from AG129 mice infected with dengue virus, serotype- 2 (S221) was assessed by plaque assay (Figure
1) and qRT-PCR (Figure 2) for the detection of viral load at various timepoints.
