A Cytopathic Effect (CPE) inhibition assay is a method used to evaluate the effectiveness of antiviral agents, such as drugs or experimental treatments, in preventing or reducing the cytopathic effects caused by viral infections in cultured cells. This assay is a critical tool in the field of virology for assessing the potential of antiviral compounds.

SMALL MOLECULE INHIBITORS TESTED AGAINST DENV-2 Y98P IN A CPE INHIBITION ASSAY

Test article (TA) 1 and TA2 are small molecule inhibitors tested against DENV-2 Y98P in Vero cells. TA1 is the better inhibitor of DENV-2 Y98P as TA1 causes 100% inhibition at the first three tested concentrations while DENV-2 Y98P inhibition is observed only at the first tested concentration of TA2. An internal control was tested in parallel

PRODUCTS TESTED AGAINST INFLUENZA A/HONG KONG/1/68 H3N2 IN A CPE INHIBITION ASSAY

TA1 and TA2 are products tested for their antiviral efficacy against IFV-A/HK/1/68 in MDCK cells. Inhibition of influenza was observed in MDCK cells treated with TA1,but was not observed in cells treated with TA2. The IBT internal control is a small molecule inhibitor of influenza that is used for assay validation.

The CPE inhibition assay is based on the principle that many viruses, when they infect host cells, cause distinct visible changes in the morphology or behavior of these cells. These changes, referred to as cytopathic effects, can include cell rounding, detachment, cell death, or the formation of characteristic inclusion bodies. Antiviral compounds can potentially interfere with viral replication and reduce these harmful effects on the host cells.

OUR EXPERTISE

APPLICATIONS OF THE CPE INHIBITION ASSAY

VIRUS IDENTIFICATION

CPE Assays are often used to identify unknown viruses by observing the distinctive changes they induce in cell cultures.

ANTIVIRAL DRUG SCREENING

Researchers use CPE Assays to test the efficacy of potential antiviral drugs. A decrease in cytopathic effects in the presence of a drug indicates its ability to inhibit viral replication.

VACCINE DEVELOPMENT

Evaluating the cytopathic effects of candidate vaccines on cell cultures helps assess their potential for protecting against viral infections.

VIRAL PATHOGENICITY

By studying the extent and nature of cytopathic effects, researchers can gain insights into the pathogenicity of different viral strains.
